ABSTRACT

BACKGROUND@#Congenital scoliosis (CS) is a complex spinal malformation of unknown etiology with abnormal bone metabolism. Fibroblast growth factor 23 (FGF23), secreted by osteoblasts and osteocytes, can inhibit bone formation and mineralization. This research aims to investigate the relationship between CS and FGF23.@*METHODS@#We collected peripheral blood from two pairs of identical twins for methylation sequencing of the target region. FGF23 mRNA levels in the peripheral blood of CS patients and age-matched controls were measured. Receiver operator characteristic (ROC) curve analyses were conducted to evaluate the specificity and sensitivity of FGF23. The expression levels of FGF23 and its downstream factors fibroblast growth factor receptor 3 (FGFr3)/tissue non-specific alkaline phosphatase (TNAP)/osteopontin (OPN) in primary osteoblasts from CS patients (CS-Ob) and controls (CT-Ob) were detected. In addition, the osteogenic abilities of FGF23-knockdown or FGF23-overexpressing Ob were examined.@*RESULTS@#DNA methylation of the FGF23 gene in CS patients was decreased compared to that of their identical twins, accompanied by increased mRNA levels. CS patients had increased peripheral blood FGF23 mRNA levels and decreased computed tomography (CT) values compared with controls. The FGF23 mRNA levels were negatively correlated with the CT value of the spine, and ROCs of FGF23 mRNA levels showed high sensitivity and specificity for CS. Additionally, significantly increased levels of FGF23, FGFr3, OPN, impaired osteogenic mineralization and lower TNAP levels were observed in CS-Ob. Moreover, FGF23 overexpression in CT-Ob increased FGFr3 and OPN levels and decreased TNAP levels, while FGF23 knockdown induced downregulation of FGFr3 and OPN but upregulation of TNAP in CS-Ob. Mineralization of CS-Ob was rescued after FGF23 knockdown.@*CONCLUSIONS@#Our results suggested increased peripheral blood FGF23 levels, decreased bone mineral density in CS patients, and a good predictive ability of CS by peripheral blood FGF23 levels. FGF23 may contribute to osteopenia in CS patients through FGFr3/TNAP / OPN pathway.

ABSTRACT

Online courses are an indispensable part of medical teaching in the new era. Online courses have good prospects, although also with certain problems in practice. As an important basic medical course, medical genetics has both basic theoretical knowledge and clinical cases, involving basic principles and the latest developments. A single online course or offline teaching model cannot meet the needs of subject development and training a new generation of medical professionals. Therefore, actively exploring the online and offline hybrid teaching model is one of the important topics in the current medical teaching reform.

ABSTRACT

Objective To develop an HPLC-MS/MS method with solid nuclear particle chromatographic column to determine 15 afrodyn drugs illegally added in sex-enhancing product. Methods The analysis was performed by an HPLC-MS/MS system, with Cortecs-C18 (150 mm × 4.6 mm, 2.7 μm) column. Multiple-reaction monitoring (MRM) was performed to identify and quantify phentolamine mesylate, noracetildenafil, acetildenafil, vardenafil, sildenafil, hydroxyhomosildenafil, homosildenafil, aminotadalafil, tadalafil, thioaildenafil, pseudovardenafil, norneosildenafil, methyltestosterone, stanozolol and danazol, which were extracted with methanol by ultrasonic. Results The linear calibration curves of 15 chemical components mentioned above were obtained at r2 ≥ 0.995 4 with better separation degree and wide linear range. The precision of the method was shown by RSD (n = 6) from six estimated values ranged from 0.9% to 3.3%. The recovery rates determined at three adding concentrations were from 88.3% to 109.5% and the RSD was from 0.9% to 5.2%. The range of limit of quantitation (LOQ) were from 0.015 μg/mL to 0.30 μg/mL and The RSD of inter-day and intra-day precision were from 0.9% to 4.2% and from 1.0% to 4.5%, respectively; Twenty batches of positive samples of 25 batches of sex-enhancing products were detected to be added with sildenafil and with an unqualified rate of 80%. Conclusion The method is specific and simple to detect that 15 afrodyn drugs were added illegally in sex-enhancing product. At the same time, the high positive detection rate should attract the attention of the department concerned, and the corresponding policies should be issued to guarantee the health and safety of the people.

ABSTRACT

BACKGROUND: Micro-arc oxidation (MAO) treatment can improve the biological properties of titanium alloy implants. Previous studies mostly focused on the evaluation of titanium alloy plate, while the effects of the MAO-modified 3D titanium scaffold on the cell growth and differentiation were rarely reported. OBJECTIVE: To investigate the effect of MAO coating on the biological performance of cells seeded onto the 3D-printed porous titanium alloy scaffold. METHODS: Rat bone marrow mesenchymal stem cells were seeded onto the MAO-modified Ti6Al4V alloy scaffolds (experimental group) and unmodified scaffolds (control group). After 4 and 7 days of culture, cell/scaffold constructs were retrieved and processed for the assessment of cell morphology by using scanning electron microscopy, cytoskeletal staining analysis and cell viability assay were also evaluated. At 4, 7 and 11 days of culture, the levels of alkaline phosphatase and osteocalcin in the cell supernatant were detected. At 1, 4, 7 and 11 days of culture, the cell proliferation rate was measured using the MTT assay. RESULTS AND CONCLUSION: (1) At 4 and 7 days of culture, live/dead staining showed that the bone marrow mesenchymal stem cells grew well on the two kinds of scaffolds. The analysis of cytoskeleton staining showed that the cytoskeleton of the experimental group was stereo and polygonal, while the cells on the scaffold surface in the control group were flat and spindle-shaped, spreading along the macro structure of the scaffolds. Under the scanning electron microscopy, the cells in the experimental group arranged closely and spread in a good condition, with interconnected lamellipodia and filopodia that firmly adhered to the scaffold surface in an anchor-shaped structure; in the control group, less filopodia interconnected, less extracellular matrix, and flat and sheet-like cells were observed. (2) With the time increase, the levels of alkaline phosphatase and osteocalcin increased gradually in both groups. The alkaline phosphatase level in the experimental group was significantly higher than that in the control group at 7 and 11 days of culture (P < 0.05), while the osteocalcin level was higher in the experimental group than the control group at 11 days of culture (P < 0.05). (3) With the prolongation of culture time, the number of cells in the two groups increased gradually. The number of cells cultured in the experimental group was significantly higher than that in the control group at 7 and 11 days of culture (P < 0.05). To conclude, the MAO-modified titanium alloy scaffold is favorable for cell adhesion, proliferation and differentiation.

ABSTRACT

Objective To evaluate the clinical effect and advantage of endovascular sclerotherapy of venous malformations in oral region using absolute ethanol under the guide of DSA.Methods The clinical data of 78 patients with venous malformation in oral region, who were indicated for ethanol sclerotherapy under DSA was retrospectively reviewed.Type classification of venous malformations was confirmed by using DSA subtraction technique during intraoperative puncture.Under general anesthesia,the malformations were punctured and then sclerosed with absolute ethanol by DSA guidance.The treatment was gone every other month,2 -6 times as a course.The length of the course of treatment depended on the type of venous malformations.Complications were also reported and analyzed. The duration of follow-up ranged from 6 to 18 months.Results Postoperative MRI showed,32 cases were cured,35 cases were significantly improved,8 cases were improved and 3 cases were invalid.There were no serious systemic complications in all cases,only 4 cases with reversible local necrosis and vesiculation.Conclusion Sclerotherapy of oral venous malformations using absolute ethanol,for its less trauma,no scar after operation,strong repeatability and exact effect,which is worthy of clinical popularization and application.

Objective To investigate the effect of lumboperitoneal (L-P) shunt and ventriculoperitoneal(V-P) shunt for trea-ting the patients with communicating hydrocephalus .Methods The databases of PubMed ,Web of Science ,Scopuss ,Karge , EBSCO+MEDLINE ,OVID ,EMBASE ,CNKI ,CBM disc databases ,Wanfang databases ,Weipu databases were retrieved by com-puter .The relevant literatures about L-P shunt and V-P shunt for treating communicating hydrocephalus included in these databases during 1990-2016 were collected and performed the meta analysis by using the STATA 12 .0 software .Results The success rate of L-P shunt in treating communicating hydrocephalus was apparently higher than that of V-P shunt(P<0 .05) .Moreover ,postopera-tive infection rate ,obstruction rate of shunt system and total postoperative complications rate in L-P shunt were apparently lower than those of V-P shunt(P<0 .05) ,However ,there was no statistical difference in shunt poor rate between L-P shunt and V-P shunt(P>0 .05) .Conclusion L-P shunt is worth recommending .But due to lower quality of the evidences ,it is needed more high quality primary studies to remedy the insufficiency of the study .

Objective To prepare a targeted ultrasound micro bubble,which carried the HSV-TK gene,and investigate the in vitro target searching ability of the micro bubbles and inhibitory effect on the proliferation of HepG2 cells.Methods Ultrasonic micro bubbles were prepared by mechanical vibration method,construction of targeted HSV-TK ultrasound micro bubbles by biotin affinity bridge construction.To detect the general characteristics of ultrasound micro bubbles,and to test its effect on the proliferation of HepG2 cells in vitro.Results HSV-TK targeted ultrasound microbubbles more gathered on the surface of HepG2 cells,through detection of PCNA and MTT,it was found that the proliferation of gene targeting microbubble group was obviously decreased,cell apoptosis increased significantly,Cells invade experiments showed that the number of cells in genetic microbubble group (22.18 ± 2.01) decreased significantly compared with the control group and the nontargeted group,can effectively inhibit the proliferation and invasive ability of HepG2 cells.Conclusion Targeted ultrasound microbubble carrying target gene have better inhibitory effect on HepG2 cells in vitro.

Objective: To develop an HPLC method with solid nuclear particle chromatographic column to determine 13 anti-diabetic drugs illegally added in health food. Methods: The analysis was performed by an HPLC system of Waters e2695, with Cortecs-C18 (100 mm × 4.6 mm, 2.7 μm) column. PDA was performed to identify and quantify tolbutamide, glibenclamide, gliclazide, glipizide, gliquidone, glimepiride, rosiglitazone, repaglinide, pioglitazone hydrochloride, metformin, phenformin, buformin hydrochloride, and glibomuride, which were extracted with methanol by ultrasonic. Results: Thirteen linear calibration curves were obtained with r2 ≥ 0.998 7. The precision of the method was shown by RSD (n = 6) ranged from 0.6% to 1.5%. The recoveries were determined at three concentrations and ranged from 95.9% to 104.7%. The ranges of LLOQ were from 1.1 μg/mL to 3.3 μg/mL and the RSDs (n = 9) of inter-day precision were from 0.5% and 1.6%. Seventeen batches in 86 antidiabetic health foods were added the chemicals with positive rate of 20%. The glibenclamide, buformin hydrochloride, rosiglitazone, glimepiride, metformin, and pioglitazone hydrochloride were determined. Conclusion: The method is specific, simple, and fast to detect 13 anti-diabetic drugs illegally added in health food.

OBJECTIVE: To develop an immunoaffinity column clean-up and high performance liquid chromatography coupled with triple quadrupole mass spectrometry(HPLC-MS/MS) method to determine aflatoxins in gelatin drugs. METHODS: The analysis was performed by an HPLC-MS/MS system with X-Brigde-C18(3.0 mm×50 mm, 3.5 μm)column. Multiple-reaction monitoring (MRM) was performed to identify and quantify aflatoxin B1, B2, G1 and G2, which were extracted from Asini Coril Colla, Cervil Cornus Colla, and Testudinis Carapacis Colla with 60% methanol solution. RESULTS: Linear calibration curves were obtained with r≥0.997 9. The precision of the method was showed by RSDs (n=6) ranging from 1.2% to 4.1%. The recoveries were determined at three concentration levels and ranged from 77.3% to 94.6%. The ranges of LOQs were from 0.5 to 0.8 μg·L-1 and the RSDs (n=9) of intra-day precision and inter-day precision were from 1.1% to 2.9% and from 1.5% to 2.8%, respectively. CONCLUSION: The method is specific, simple and rapid to detect aflatoxins in gelatin drugs.

ABSTRACT

Objective Cancer, a disease induced by abnormally regulated cell growth and apoptosis, is imposing a global threat to human health.This study was to explore the effects of Chinese herbal extracts ( CHE) in inducing the apoptosis and inhibiting the proliferation of human lung cancer cells. Methods Human lung cancer A549 cells were divided into a negative control, a high-dose CHE (680 ng/mL), a medium-dose CHE (340 ng/mL), and a low-dose CHE (170 ng/mL) group.The inhibitory effect of CHE on the proliferation of the lung cancer cells was detected by CCK8 and LDH assays, the apoptosis of the cells was assessed by fluorescence microscopy and flow cytometry, and the expressions of hTERT mRNA, cleaved caspase-3 and cleaved PARP were deter-mined by RT-PCR and Western blot. Results CHE inhibited the proliferation of the A549 cells with an IC50 value of 510 ng/mL. Treatment with high-dose CHE for 48 hours significantly suppressed the proliferation of the cells, induced the release of LDH, and promo-ted the apoptosis of the cells by 72.3%.RT-PCR and Western blot showed that 24-hour treatment with medium-dose CHE reduced the expression of hTERT mRNA by 4 times that of the negative control and up-regulated the expressions of cleaved caspase-3 and cleaved PARP. Conclusion Chinese herbal extracts can induce cell apoptosis by decreasing the expression of hTERT mRNA and increasing those of the cleaved caspase 3 and cleaved PARP proteins.

ABSTRACT

Objective To probe the application value of “double low”CTU method in diagnosis of urinary system malformation in children.Methods Patients with urinary system malformation were screened by ultrasound,and renal calculi,tumor and tumor-like lesions were excluded.Double phase contrast media injection (iodixanol contrast media 270 mg I/mL)was used in one phase scan-ning,and the cortex-secretory phase images were obtained.The images were reviewed and evaluated independently by two readers. Results The coincidence rate of diagnosis was 100% with well images quality.Conclusion CTU is an important tool for the diagno-sis of urinary tract malformations in children.The “double low”method with lower radiation dose and contrast media quantity may ensure the quality of diagnosis.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the dynamic changes of transforming growth factor-α (TGF-α) and transforming growth factor-β1 (TGF-β1) of liver cirrhosis induced by multiple pathogenic factors in rats.</p><p><b>METHODS</b>Animals in the cirrhosis group were fed a mixture of maize flour, lard, cholesterol and alcohol plus subcutaneously injection with carbon tetrachloride (CCl₄), the CCl₄(0.5 ml/100 g · w) was injected at the first day of experiment and the 40% CCl₄oil solution (0.3 ml /100 g · w) was injected at an interval of three days. The thirty-six male SD rats were randomly divided into liver cirrhosis group of the 4th, 6th and 8 th week, and normal control group of the 4th, 6th and 8th week. The contents of alanine transferase (ALT), endotoxin, tumor necrosis factor-α (TNF-α) and homocysteine (Hcy) in plasma were evaluated. Histopathological changes of the liver were observed under microscope with the staining of HE. The expressions of TGF-α and TGF-β1 were analyzed by the method of immunohistochemistry.</p><p><b>RESULTS</b>Compared with the corresponding normal control group, the levels of ALT, endotoxin, TNF-α and Hcy in plasma were gradually significantly increased in liver cirrhosis group of the 4th, 6th and 8th week (P < 0.05); the expression of TGF-α in the liver tissues was significantly increased at the 4th week (P < 0.05); the expression of TGF-β1 in the liver tissues was gradually significantly increased in every model group (P < 0.05).</p><p><b>CONCLUSION</b>In the formation process of cirrhosis, the expression of TGF-α was increased in liver of cirrhosis group at the 4th week, and later it was suppressed; the expression of TGF-β1 was continuously increased. The characteristic dynamic changes of TGF-α and TGF-β1 might be related to sustained endotoxemia, the high level of TNF-α and hyperhomocysteinemia.</p>

BACKGROUND: Psoriasis is an autoimmune disease that is caused by a shift in the Th1/Th2 balance toward Th1-dominant immunity. It has been established as an effective treatment to counteract psoriasis by subcutaneous injection of recombinant interleukin (IL)-4, and IL-4 gene therapy by topical transdermal penetration has shown its antipsoriatic effect in mice. Retinoic acid (RA) and dimethylsulfoxide can increase the efficiency of gene transfection in the topical transdermal delivery system. OBJECTIVE: We investigated whether RA could improve anti-psoriasis efficiency using IL-4 expression plasmid pORF-mIL-4 (pIL-4) via transdermal delivery system in K14-vascular endothelial growth (K14-VEGF) factor transgenic mice. METHODS: After pretreatment with RA, plasmid pIL-4 in 10% dimethylsulfoxide was applied to the ear skin by topical transdermal penetration. Hematoxylin- eosin staining and immunohistochemistry were performed with ear samples to evaluate anti-psoriasis efficiency in mice. RESULTS: The psoriasis pathological features were relieved and psoriasis-associated factors were significantly reduced. CONCLUSION: Our results reveal that topical application of pIL-4 in dimethylsulfoxide by transdermal delivery with RA pretreatment can improve psoriasis significantly.

ABSTRACT

Objective To study the effect of postoperative ileus of laparoscopic surgery versus open surgery for colorectal canc-er .Methods 220 patients of colorectal cancer were selected from October 2009 to June 2012 .The patients were randomly divided into the observation group(n=110 cases) and the control group(n=110 cases) .The patients of the observation group were treated with laparoscopic surgery ,and the patients of the control group were treated with conventional open surgery .The postoperative anal exhaust time and hospital stay time ,the incidence of postoperative ileus of the two groups were observed and compared .Results The postoperative anal exhaust time and hospital stay time of the patients after radical resection of left colon ,right colon and rectal cancer of the observation group were significantly less than that of the control group (P<0 .05) .The incidence of postoperative ileus of the observation group was 5 .45% (6/110 ) ,significantly lower than 11 .82% (13/110 ) of the control group ( P< 0 .05 ) . Conclusion Laparoscopic colorectal surgery is a minimally invasive surgery ,the patient′s trauma is smaller ,the postoperative re-covery is rapidly .The incidence of intestinal obstruction is smaller than that of the conventional open surgery .

Objective: To develop a UPLC-MS/MS method for determination of eight antirheumatic cconstituents illegally added in Chinese patent medicine preparation. Methods: The analysis was performed by a UPLC-MS/MS system of Waters Acquity UPLC/ Quattro Premier, with BEH-C18 (100 mm × 2.1 mm, 1.7 μm) column. Multiple-reaction monitoring (MRM) was performed to identify and quantify hydrocortisone, dexamethasone, prednisone 21-acetate, 4-acetamidophenol, phenylbutazone, naproxen, piroxicam, and trimethoprim, which were extracted with methanol by ultrasonic method. Results: Eight linear calibration curves were obtained with r2 ≥ 0.998 9. The precision of the method was shown by RSD (n = 6) ranged from 1.2% to 3.5%. The recoveries were determinated at three concentration and ranged from 95.4% to 104.9%. The ranges of LLOQ were from 0.4 to 4.9 μg/mL and the RSDs (n = 9) of intra-day precision and inter-day precision were from 0.6% to 2.7% and from 0.9% to 2.9%. ABSTRACT

<p><b>OBJECTIVE</b>To explore the mechanism of tanshinol on alleviate the inflammatory injury of lung tissue in rat hepatopulmonary syndrome (HPS).</p><p><b>METHODS</b>SD rats were randomly divided into normal control group (n = 8), hepatopulmonary syndrome (HPS) group (n = 11) and tanshinol intervention group (n = 9). HE staining was used to observe the histopathology changes of pulmonary and hepatic tissues, and to count the number of macrophages in lung tissues. The activity of alanine transferase (ALT) and concentrations of endotoxin, tumor necrosis factor-a (TNF-alpha) and homocystein (Hcy) in plasma were detected. The concentrations of TNF-alpha, nitric oxide (NO) and malondialdehyde (MDA) and the activity of inducible nitric oxide synthase (iNOS) in the lung tissues were measured, respectively.</p><p><b>RESULTS</b>Thickened alveolar septum and increased macrophages were observed in lungs in HPS rat. After administered with tanshinol, the pulmonary pathological changes were alleviated and the number of macrophages in lung tissue was decreased compared with HPS group. The activity of ALT and the concentrations of endotoxin, TNF-alpha and Hcy in plasma ,and TNF-alpha, iNOS, NO and MDA in lung tissue in HPS group were higher than those of normal control group; meanwhile, those tanshinol group were less those that of HPS group.</p><p><b>CONCLUSION</b>Tanshinol may play an important role in delaying the development of HPS through protecting liver or directly antagonizing the effect of intestinal endotoxemia so as to alleviate the inflammatory reaction in lung tissue.</p>

ABSTRACT

Objective This study aimed to investigate whether platelet-derived growth factor CC could enhance resolution ,or-ganization and recanalization of venous thrombus .Methods The experimental models of deep vein thrombosis in rats were induced as previously described and modified .The rats were randomly divided into four groups according to the dose of PDGF-CC(Group A :0 ng/100 μL ;Group B :100 ng/100 μL ;Group C :200 ng/100 μL and Group D :500 ng/100 μL) .The samples were obtained at 7 days after operation and the expressions of protein VEGF were detected by Western blot analysis ,respectively .Immunohistochemi-cal staining was used to observe the types of the positive cells expressing vWF in thrombus sections .HE staining and Masson stai-ning were used to analyze the area of resolution and organization in venous thrombi .The capillary number was analyzed quantitative-ly by high-power microscope .The area of thrombus and collagen fiber in each section were measured and analyzed with image analy-sis software .Results Animal models of venous thrombogenesis were performed successfully .The data of Western blot analysis demonstrated that the expressions of VEGF protein were significantly increased in group C and D than group B and A ,which were no statistical significance between group C and group D at day 7 after operation .The positive cells of expressing vWF mostly located in the thrombus border and venous wall and the types of the positive cells by immunohistochemical staining were ECs .The resolu-tion rates ,organization rates and recanalization capillary numbers were significantly higher in group C and D than those in group B and A ,and those in group B were higher than those in group A .Conclusion PDGF-CC could enhance resolution ,organization and recanalization of vein thrombi effectively through therapeutic angiogenesis pathway ,which provides a novel strategy for gene thera-py of venous thromboembolism disease .

ABSTRACT

Patients with late-stage cancer commonly have distant lymph node metastasis; however, poor health often contraindicates surgical treatment. Although the quality of life and overall survival for these patients are low, there is neither a consensus nor a guide for treatment. Ablation technique and surrounding tissue damage are two possible reasons for limited study of radiofrequency ablation in patients with superficial distant lymph node metastasis. Here, we report two patients treated successfully with ultrasound-guided radiofrequency ablation for superficial distant lymph node metastasis. In these patients, deionized water was injected to the surrounding tissues of the lymph node to decrease heat injury. Results from these patients suggest that radiofrequency ablation may play an important role in the treatment of patients with distant lymph node metastasis.

ABSTRACT

@#Objective To observe the effects of the home tele-rehabilitation guidance on activities of daily living and motor function in patients after cerebral infarction. Methods 101 cases with cerebral infarction at recovery stage were divided into control (n=50) and rehabilitation (n=51) group. The rehabilitation group received home tele-rehabilitation guidance. They were assessed with the Barthel index (BI) and the simplified Fugl-Meyer assessment (FMA) before and 3 month after treatment. Results There is no significant difference in both the BI and FMA between these groups before treatment, and the rehabilitation group improved more after treatment (P<0.05). ConclusionHome tele-rehabilitation guidance can facilitate the recovery of activities of daily living and motor function in cerebral infarction patients.
